Rybelsus (semaglutide) has been associated with a potential risk of thyroid tumors, including medullary thyroid carcinoma (MTC). This concern is based on studies conducted in rodents, where semaglutide caused a dose-dependent increase in the incidence of thyroid C-cell tumors. However, it is not known if Rybelsus causes these tumors in humans.

Due to this potential risk, Rybelsus is contraindicated in individuals with a personal or family history of MTC or those with Multiple Endocrine Neoplasia syndrome type 2 (MEN 2).
